以4~6个月的实生苗茎段为外植体,研究了离体条件下培养基及激素对栓皮栎器官发生和植株再生的影响。结果表明:初代培养选用低盐培养基WPM、BTM和GD,附加0.2mg·L-16-BA,丛生芽较多,茎粗壮,生长势健壮,均好于高盐培养基MS。继代培养时,以BTM为基本培养基,添加0.2~0.6mg·L-16-BA有利于茎芽增殖和生长,当6-BA浓度为0.8~1.0mg·L-1时,不利于茎芽伸长。以WPM为基本培养基,附加NAA0.1mg·L-1和IBA0.25mg·L-1时,生根率高,根系发育好。  相似文献

Cladoptosis, the abscission of twigs, is the main mechanism of changes in crown structure in senescing pedunculate oak (Quercus robur L.). We tested the hypotheses that abscission zones in nodes of old pedunculate oak trees reduce leaf-specific hydraulic conductance of shoots and thereby limit the stomatal conductance and assimilation.Hydraulic conductance and leaf-specific hydraulic conductance, measured with a high pressure flowmeter in 0.5–1.5 m long shoots, were significantly lower in shoots of low vigour compared to vigorous growing shoots in a 165-years-old stand in the southeast of Germany. Two types of bottlenecks to water transport could be identified in shoots of old oak trees, namely nodes and abscission zones. In young twigs, vessel diameter and vessel density in nodes with abscission zones were significantly reduced compared with internodes. In nodes without abscission zones, vessel density was significantly reduced. The reduction of hydraulic conductance was especially severe in the smallest and youngest shoots with diameters less than 2 mm. Internodes of 1–5 mm sapwood diameter had an average hydraulic conductance of 7.13×10−6±0.2×10−6 kg s−1 m−1 MPa−1, compared to 4.54×10−6±0.3×10−6 kg s−1 m−1 MPa−1 in those with nodes.Maximum stomatal conductance and maximum net assimilation rate increased significantly with hydraulic conductance and leaf-specific hydraulic conductance. Maximum rate of net photosynthesis Amax of the most vigorous shoots (VC0) (7.34±0.55 μmol m−2 s−1) was significantly higher (P<0.001) than in shoots of other vigour classes (5.97±0.28 μmol m−2 s−1). Our data support the hypothesis that the changes in shoot and consequently crown architecture that are observed in ageing and declining trees can limit photosynthesis by reducing shoot hydraulic conductance. Abscission zones increase the hydraulic disadvantage of less vigorous compared to vigorously growing twigs. Cladoptosis might serve as a mechanism of selection between twigs of different efficiency.  相似文献

Galphimia glauca produces compounds denominated galphimines (galphimine‐A, galphimine‐B and galphimine‐E). Due to their important anxiolytic activity, we initiated in vitro cultures of the species with the purpose of developing a biotechnological process for obtaining galphimines. In this work, we stimulated the biosynthesis and excretion of galphimines with two‐phase batch‐type cell suspension cultures of G. glauca. The effect of nutritional variation and the 2,4‐dichlorophenoxy acetic acid added to Murashige & Skoog(MS) culture medium was evaluated. Later, we evaluated the effect of the stimulation with calcium and methyl jasmonate (MeJ). The greatest production of galphimine‐B (3.39 × 10?5 g/L day?1) was obtained on day 40 of kinetics, and induced by a treatment containing concentrations of nitrates and phosphate that are double of those normally used in MS medium, without sucrose but with added 2,4‐dichlorophenoxy acetic acid (4 mg/L). Time of galphimine‐B biosynthesis diminished due to the effect of MeJ in combination with calcium, and induced the excretion (100%) of galphimine‐B (6.35 × 10?5 g/L day?1) into the culture medium. Thus, the use of calcium and MeJ comprises a viable alternative to stimulate the production and excretion of galphimine‐B and galphimine‐A in batch‐type cultures of G. glauca in modified MS medium. Once optimized, the production of the anxiolytic compounds can be scaled up to the industrial level.  相似文献

We measured acorn production by four species of oaks in central Minnesota over a 17-year period with the goal of understanding the proximate drivers of masting behavior. All four species exhibited significant annual variation and within-population synchrony of acorn production, although masting behavior was more pronounced in the two species that require 1 year to develop acorns (‘1-year’ species) than the two species that require 2 years (‘2-year’ species). There was also strong synchrony between species that require the same number of years to mature acorns, but not between species requiring different numbers of years. Acorn production by three of the four species correlated with spring or summer conditions, while no significant environmental correlate of acorn production was detected for the fourth species. Acorn production by none of the four species correlated significantly with variables calculated from the differences in weather conditions from 1 year to the next. These results, combined with prior studies of oaks, suggest that environmental conditions during key periods of acorn development frequently correlate with acorn production, as expected if such factors bear a direct mechanistic relationship to seed production. On the other hand, the environmental factors involve vary greatly both among species and even among populations of the same species, a result consistent with the hypothesis that environmental correlates are simply cues used by plants to synchronize reproductive investment. In either case, our results do not support the recent proposition that variables based on differences in environmental conditions from 1 year to the next serve as a general cue for masting behavior.  相似文献

Background and Aims

Hybridizing species such as oaks may provide a model to study the role of selection in speciation with gene flow. Discrete species'' identities and different adaptations are maintained among closely related oak species despite recurrent gene flow. This is probably due to ecologically mediated selection at a few key genes or genomic regions. Neutrality tests can be applied to identify so-called outlier loci, which demonstrate locus-specific signatures of divergent selection and are candidate genes for further study.

Methods

Thirty-six genic microsatellite markers, some with putative functions in flowering time and drought tolerance, and eight non-genic microsatellite markers were screened in two population pairs (n = 160) of the interfertile species Quercus rubra and Q. ellipsoidalis, which are characterized by contrasting adaptations to drought. Putative outliers were then tested in additional population pairs from two different geographic regions (n = 159) to support further their potential role in adaptive divergence.

Key Results

A marker located in the coding sequence of a putative CONSTANS-like (COL) gene was repeatedly identified as under strong divergent selection across all three geographically disjunct population pairs. COL genes are involved in the photoperiodic control of growth and development and are implicated in the regulation of flowering time.

Conclusions

The location of the polymorphism in the Quercus COL gene and given the potential role of COL genes in adaptive divergence and reproductive isolation makes this a promising candidate speciation gene. Further investigation of the phenological characteristics of both species and flowering time pathway genes is suggested in order to elucidate the importance of phenology genes for the maintenance of species integrity. Next-generation sequencing in multiple population pairs in combination with high-density genetic linkage maps could reveal the genome-wide distribution of outlier genes and their potential role in reproductive isolation between these species.  相似文献

Ectomycorrhizal fungi are major ecological players in temperate forests, but they are rarely used in measures of forest condition because large‐scale, high‐resolution, standardized and replicated belowground data are scarce. We carried out an analysis of ectomycorrhizas at 22 intensively monitored long‐term oak plots, across nine European countries, covering complex natural and anthropogenic environmental gradients. We found that at large scales, mycorrhizal richness and evenness declined with decreasing soil pH and root density, and with increasing atmospheric nitrogen deposition. Shifts in mycorrhizas with different functional traits were detected; mycorrhizas with structures specialized for long‐distance transport related differently to most environmental variables than those without. The dominant oak‐specialist Lactarius quietus, with limited soil exploration abilities, responds positively to increasing nitrogen inputs and decreasing pH. In contrast, Tricholoma, Cortinarius and Piloderma species, with medium‐distance soil exploration abilities, show a consistently negative response. We also determined nitrogen critical loads for moderate (9.5–13.5 kg N/ha/year) and drastic (17 kg N/ha/year) changes in belowground mycorrhizal root communities in temperate oak forests. Overall, we generated the first baseline data for ectomycorrhizal fungi in the oak forests sampled, identified nitrogen pollution as one of their major drivers at large scales and revealed fungi that individually and/or in combination with others can be used as belowground indicators of environmental characteristics.  相似文献

参照CTFS技术规范对灵空山自然保护区油松(Pinus tabuliformis)-辽东栎(Quercus wutaishanica)林的乔木进行每木调查的基础上分析了不同乔木树种的群落学特征及空间分布格局。结果表明,4hm2样地中共有乔木树种25种,总计8210株。隶属于12科18属,种的区系以东亚、温带亚洲及中国特有分布类型为主。油松、辽东栎是群落中的共优种,个体占总个体的86.81%,杜梨(Pyrus betulifolia)、白蜡树(Fraxinus chinensis)、漆树(Toxicodendron vernicifluum)为群落中主要的伴生种。优势种的径级结构与所有乔木个体的径级结构相似,近似于倒"J"形,以小径级个体居多,林下更新良好。主要树种的空间分布格局在小尺度上表现为聚集分布。油松小径级个体在20 m以内的尺度上呈聚集分布,当空间尺度超过60 m时表现为随机分布,大径级个体在所有尺度上均呈随机分布;辽东栎不同径级个体在小尺度范围内呈现聚集分布,随着尺度增大由随机分布趋于均匀分布。主要树种除漆树外在坡度较缓的生境下分布密度大。灵空山自然保护区油松-辽东栎林中油松种群优势地位更显著,各个乔木种群处于相对稳定状态。  相似文献

Meloidogyne christiei n. sp. is described and illustrated from turkey oak (Quercus laevis) in Sanlando Park, Altamonte Springs, Florida. This new nematode species has a distinctive perineal pattern commonly with a high, squarish arch and coarse broken striae which tend to diverge at various angles, especially in and above the anal area. Female labial disc is indented, forming four points or prongs, unlike other species. Eggs are deposited inside the gall in a tubular, coiled manner. Vaginal muscles are exceptionally prominent and dense. SEM observations provided further detail of the perineal pattern and details of the head of females, males, and second-stage juveniles. Galls on the root commonly occur singly, but sometimes in small clusters, and appear as discrete nodules on the side of the root and without adjacent swelling. In general, only one female is found in each gall but occasionally two are present. In greenhouse tests, citrus, tobacco, cotton, pepper, watermelon, peanut, and tomato were not hosts. This nematode occurs throughout central Florida commonly on Q. laevis, the only known host.  相似文献

Abstract. On the granite outcrops of the southeastern United States, soil accumulates in shallow depressions on the rock surface. A specific sequence of vascular plants characterizes the temporal development of these systems. The edaphic end point of the succession is apparently attained with a herb-shrub-tree stage with Pinus taeda as the dominant tree. We studied the characteristics of this stage and the population structure of P. taeda on outcrop islands in order to specify the successional status of the species in this habitat. We compared the radial growth pattern of outcrop and Piedmont populations of P. taeda, and of two outcrop sub-populations. We checked whether trees on outcrops experience more limiting conditions than trees on the Piedmont, and studied the recent change in the relationship between growth and stress (e.g. drought and atmospheric deposition) reported for loblolly pine in the southeastern United States. We also attempted to identify the climatic variables most critical for tree growth on outcrops. On outcrop soil islands, P. taeda maintains populations that are of irregular age distribution, possibly in response to irregular recruitment and survival. There are no signs of loblolly pine replacement by hardwood species on any of the islands studied, although an understory of shrubby oak appears to characterize larger and deeper-soil islands. Although trees on the Piedmont were growing at a higher rate than those on the outcrop in the first part of the 1950–1988 period, their growth declined at a higher rate than that of trees on the outcrop. As a result, at the end of the period considered (1950–1988), the differences in radial growth between outcrop and Piedmont populations were relatively small. On the outcrop, trees < 22 yr old in 1989 were growing at a rate somewhat lower than that of trees of similar age, 40 yr ago. Differences were apparent in the initial growth patterns between the two outcrop subpopulations (1989 and 1949 stems), and these could have resulted from differences in competition regime, stress or climatic conditions, or a combination thereof. However, a repeated-measures ANOVA failed to reveal a significant recent decrease in the radial growth of loblolly pine in the system studied. The identification of numerous signature years (years with ≥ 80 % of the trees with similar increasing or decreasing trend in their radial growth) suggests that similar environmental variables control the growth of loblolly pine in both outcrop and Piedmont habitats. High temperature and low precipitation in the first part of the summer (June-July) seem to limit radial growth. Those pines growing on outcrop soil islands, however, appear more sensitive to climatic fluctuations.  相似文献
